Patrick Barlow a joué dans plusieurs longs métrages dont LE JOURNAL DE BRIDGET JONES de Sharon Maguire, THAT GIRL FROM RIO de Christopher Monger, SHAKESPEARE IN LOVE de John Madden ou COUP DE FOUDRE À NOTTING HILL de Roger Michell.
À la télévision, il a été l'interprète de plusieurs épisodes des très populaires séries «French and Saunders» et «Absolutely Fabulous», ainsi que de «Victoria Wood As Seen on TV», et «A Bit of Fry and Laurie». Il a également joué dans «Murder in Suburbia», «Goodbye Mr Steadman», «Is It Legal», «The True Adventures of Christopher Columbus», «Van Gogh», et «The Growing Pains of Adrian Mole».
Côté théâtre, il s'est produit à Londres dans «The Wonders of Sex» au National Theatre et «The Messiah» au Bush Theatre. Il a joué aussi «Love Upon the Throne» au Comedy Theatre, à Edimbourg et au Festival de Berlin, «Wind in the Willows» au Royal National, «Shakespea-re, the Truth» un one-man show au Théâtre National de Berlin, et «Loot» au Royal Exchange. Il a par ailleurs joué régulièrement sur BBC Radio 4 et a écrit et réalisé divers projets pour la télévision.
Le 8 février 2006 il est à l'affiche de Nanny MacPhee de Kirk Jones.
